Here, we developed P-BEST, a method for Pooling-Based Efficient SARS-CoV-2 Testing, using a single-stage nonadaptive group-testing approach, which significantly reduces the number of tests required to identify all positive subjects within a large set of samples. PCR results for each of the pools are provided to the detection algorithm (see Methods), which identifies all positive carriers without the need for an additional testing stage (Fig. 1 , A to C). RNA was extracted from each single sample and pools and was then tested for SARS-CoV-2 using the Seegene COVID-19 diagnostic kit.
