Initially, the vaccinia virus-based reverse genetic system was established for the generation of recombinant human coronavirus 229E. This section describes the preparation of purified vaccinia virus DNA that can be used: (i) for the in vitro ligation with the assembled coronavirus full-length cDNA (see Section 3.1.4), and (ii) as template for in vitro transcription reactions (see Section 3.3.1). First, a fulllength coronavirus RNA is produced using the genomic DNA of a vaccinia virus containing the full-length coronavirus cDNA insert as a template for in vitro transcription. 1.
