An RNA guanylyltransferase (GTase) then catalyzes a two-step reaction in which it initially utilizes GTP as a substrate to form a covalent enzyme-GMP (EpG) intermediate, with the concomitant release of pyrophosphate (PPi). Since ribavirin triphosphate and MZP share functional similarities, we investigated the attractive possibility that MZP could also inhibit the GTase activity of the human RNA capping enzyme (HCE).
