They had nucleotide sequence identities of 97-98% and amino acid sequence identities 96-98% in common with Chinese IBVs (strain A2, SH and QXIBV). Viral RNA was extracted by using Viral Nucleic Acid Extraction Kit (Real Biotech, Taiwan) following the manufacturer's instructions directly from the supernatants of 10% w/v sample suspensions and from the allantoic fluid of embryonated chicken eggs used for virus isolation. For virus isolation, the supernatants of IBV-positive samples determined by RT-PCR were inoculated into 10-day-old embryonated chicken eggs. The results from both S1 gene comparison and phylogenetic analysis showed that IBV isolates in group I had a distant relation to vaccine strains used in Thailand including Ma5, H120, M41, and Connecticut.
