To construct a C-terminal GFP fusion of the frameshifted protein, the 535 bp 39-end fragment of the 39 ORF of the PaYIP3 gene was PCR-amplified using the orf39w-bgl2 (59-GAA-GATCTGGCTCTAGTGCATCCAGGACAC-39) and orf39c-sma1 (59-TTTCCCGGGTCGCTTTCCTTCTAGCAGTACC-39) oligonucleotides, containing, respectively, the BglII and SmaI sites (underlined in the sequence). The PaYIP3 c corrected allele was created by inserting a C in codon nu168, which is just upstream of the frameshift site, and produced the 61.6 kDa polypeptide resulting from the fusion of the proteins produced by the 39 and 59 ORFs.
